Diagnostic value of aldolase A and cytokeratin19for ovine pulmonary adenocarcinoma
Ovine pulmonary adenomatosis (OPA) is a contagious and experimentally induced lung cancer of sheep,resembling human bronchioloalveolar carcinoma (BAC).A beta-retrovirus,called jaagsiekte sheep retrovirus (JSRV),is the pathogenesis of OPA which is originating from typeⅡpneumocytes and Clara cells.JSRV particles or antigens have never been detected in the tissues of naturally affected s heep.To identify the presence of tumor metastases,a histopathological diagnosis was performed on six sheep with typical clinical manifestations.Different analytical methods were included in this retrospective study,aiming to confirm the different expression of Aldolase A and cytokeratin 19 in OPA,which were significant markers of epithelial tumor cells.Results displayed,Aldolase A expressed 1.98-fold,2.0-fold and 1.45-fold in OPA by the analysis of immunohistochemistry,western blotting and real-time quantitative fluorescent PCR,respectively.Similarly,cytokeratin 19 expressed 2.39-fold,2.4-fold and 1.70-fold in OPA.Here,we reported that the two proteins were highly expressed in the epithelial cells of bronchioles,type Ⅱ pneumocytes and tumor cells of OPA,as well as expressed in the epithelial cells of bronchioles and typeⅡpneumocytes in non-neoplastic lung sections.These data suggest that Aldolase A and cytokeratin 19 could be potential markers for comprehensive studies to investigate the presence of neoplastic cells in OPA.The hypothesis that the depletion of Aldolase A and cytokeratin 19 in OPA tumor cells which belong to the sheep of different ages and development phases alters the cell viability and tumorigenesis could be further studied fo r the drug development of therapeutic target.
