The free-exploratory paradigm as a model of trait anxiety in Kunming mice: test-retest reliability
The free exploratory paradigm has been proposed as a model of trait anxiety.By definition,trait anxiety does not vary from moment to moment,therefore any model used to evaluate it needs to give the same results in sequential trials.The aim of this study was to evaluate the test-retest reliability of free-exploratory paradigm in mice.The behavior of drug-naive,adult,male,Kunming mice was measured in the free-exploratory apparatus on two occasions,either one week apart (Experiment Ι) or four weeks apart (Experiment Π).The following parameters were evaluated: percentage of time spent in the novel compartment (Ntime%);percentage of novel compartment units visited (Ncross%);percentage of number of rears in the novel compartment (Nrear%);and attempts to enter the novel compartment (Attempts).Subsequently,the intraclass correlation coefficient (ICC) and the Kappa index (κ) were calculated for each of these parameters.Analysis of the parameter Nrear% in Experiment Ι showed the acceptable values of ICC (0.755) and κ (0.520),respectively,indicating good and fair reliability with a one-week inter-trial interval.While in Experiment Π,both ICC and κ of all the parameters failed to show a reasonable test/retest concordance,suggesting that with a four-week inter-trial interval,the reliability may diminish,or even disappear.However,our results still bring further support to free-exploratory paradigm as an animal model of trait anxiety.
