UPLC-ESI-MS analysis of You Gui Yin and its pharmacokinetics after oral administration to rats

  Objective: To develop a sensitive, specific and accurate ultra performance liquid chromatography-electronic spray ion-mass spectrometry (UPLC-ESI-MS) method and apply it for simultaneous quantitation and pharmacokinetics of six Aconitum alkaloids, i.e.aconitine (AC), hypaconitine (HA), mesaconitine (MA), benzoylaconine (BAC), benzoylhypaconine (BHA), benzoylmesaconine (BMA) in the plasma of rat after oral administration YGY.Methods: An ACQUITY UPLCTM BEH C18 column (2.1×100 mm, 1.7 μm) was used to separate the six compounds and internal standard (IS) by gradient mode with mobile phase consisting of water containing 0.1% formic acid and 2mmol/L ammonium formate and methanol containing 0.1% formic acid, at a flow rate of 0.20 mL/min with a column temperature of 40℃.UPLC-ESI-MS system coupled was performed in positive mode.Results: For the method validation, the calibration curves were linear (r>0.992) and the limit of quantification was all less than 0.1 ng/mL.The within day precisions (RSD) were all <15%, accuracies (RE) were within the range of± 15%, and recoveries ranged from 70.37~99.3%.Conclusion: The validated method was applied to simultaneous quantitation and successfully used for the first time for the pharmacokinetic evaluation of the six Aconitum alkaloids in plasma of rat after oral administration YGY.This method has high sensitivity and specificity, it could be beneficial for the application of clinical therapy.
