PP2A (PR65) in silver carp: cDNA clone and expression analysis
In the present study, the full-length sequence of protein phosphatases 2A subunit A (PP2A-A) cDNA was cloned, sequenced, and the constitutive expression of PP2A-A was also analyzed in silver carp.Moreover, the transcription of PP2A-A in the fish liver, kidney, and spleen after microcystins (MCs)-exposure were also determined in this study.The results reveal that the full length ofPP24-A cDNA is 2 833 base pair (bp) long and contains an open reading frame of 1 767 bp encoding a protein of 589 amino acids with an estimated molecular weight of 65.47 kDa and an isoelectric point of 4.95.Sequence analysis reveals that PP2A is highly conserved in fish.Furthermore, the results of quantitative real-time PCR reveal that PP2A-A is constitutively expressed in all tissues of silver carp, but mainly in brain, liver and heart.In addition, we also found that 50 or 200 μg kg-1 of crude MCs exposure remarkably down-regulated the transcription of PP2A-4 in the liver, kidney, and spleen of silver carp, suggesting that PP2A-A may be involved in MC-hepatotoxicity in silver carp.
