高温对小胸鳖甲、光滑鳖甲和黄粉虫抗冻蛋白表达的影响
The desert beetles, Microdera puuctipeuuis and Auatolica polita, are special Tenebrionidae desert insects in Gurbantonggut Desert in Central Asia. Gurbantunggut desert climate characterized by extremes oftypical temperate continental arid climate, in large temperature difference between day and night, the two insectscan well adapted to the low environmental temperature and drastically temperature changes. Teuebrio ruolitor isone of the most important pests on stored grain products, and is popular as a model species for beetles because it isrelatively easy to maintain cultures of them under laboratory conditions. Long-term breeding makes temperaturesensitivity of T ruolitor weaker than beetles survived in the wild. It is well known that antifreeze proteins play animportant role in protecting poikilothermic organisms from freezing. However, the transcripts of antifreeze proteingenes, P如afps and Apafps, were observed in the desert beetle M.puuctipeuuis and A.polita in summer, whichimplies that a novel function may exist in the antifreeze proteins from these insects. This experiment studiedexpression profiled of antifreeze protein genes in different temperatures by the fluorescence quantitative PCR. Theresults showed that Mpafp transcription level was higher in spring and winter, lower in summer, expressingabundantly after short-term high and low temperature stress and the difference in temperature stress. The researchfound that two Apafp genes variation was not uniform throughout the year with different seasonal expression ofantifreeze protein gene. Apafp914 expressed highly in late fall, whereas Apafp752 expressed highly in spring. Inthe three temperature differential treatment the expression of Truafp433 significant increased, in addition, themRNA level of TruaJ沙433 could be 13 fold of the control value at 42 0C high temperature stress, illuminated hightemperature induced plenty of the expression of Truafp gene in T.ruolitor. This study suggests that insect antifreezeprotein could be induced by heat and may be involved in thermal protection and water conservation. It is possiblethat at high temperatures they are chaperones for heat labile proteins.
