Experimental Studies of SVF on labeling with DiI , multilineage differentiation, the cellular compatibility of type Ⅰ collagen scaffold in vitro
Objective To study the advantage of SVF cells as a kind of ideal tissue engineering seed cells.Methods After isolating(0.125% collagenase type Ⅰ), SVF cells was labeled with DiI in vitro.SVF was induced to adipogenic, osteogenic and chondrogenic differentiation with the corresponding inductive culture medium.It was stained by oil O, alizarin red and alcian blue correspondingly.SVF cells was co-combined with type Ⅰ collagen scaffold.The adhesion ratio of the SVF was determined by cell count method.The morphological features and function of the SVF were observed by invert optical microscope and scanning electron microscope.Result we could get a lot of SVF cells quickly with 0.125% type Ⅰ collagenase.DiI is a kind of ideal fluorsecent dye for SVF.SVF cells could be induced to adipogenic, osteogenic and chondrogenic differentiation with the contesponding inductive culture medium.SVF cells were able to attach, grow and proliferate well on the scaffolds.Adhesion rate of SVF was (92.596±2.268)%, while that of DII labeling group was (92.488±2.262)% (P =0.964).Conclusion SVF cells could be obtained quickly with 0.125% type Ⅰ collagenase.It could be induced to adipogenic, osteogenic and chondrogenic differentiation.Collagen Ⅰ scaffold exhibits excellent cellular compatibility to SVF cells.SVF cells is a kind of ideal seed cells which has a bright future in clinical practice.
