BuShenYiQi(BSYQ) Granule Inhibits Atopic Dermatitis(AD) via a Differential Expression of Peripheral and Central Hypothalamic-Pituitary-Adrenal(HPA) Tone

  Background: dysfunction of central and peripheral Hypothalamic-Pituitary-Adrenal (HPA) axis contribute to the insufficient and insensitive of endogenous glucocorticoid which is the important pathogenesis of atopic dermatitis(AD);previous study found that Bushenyiqi Chinese herbs could improve HPA axis function.Methods: The role of peripheral and central HPA axis in the pathogenesis of AD were investigated by ovalbumin(OVA) induced AD and normal BALB/c mice,meanwhile,the effect and mechanism of BuShenYiQi granule(BSYQ) on AD mice were explored.Serum corticosterone (CORT),adrenocorticotropic hormone (ACTH),corticotropin-releasing hormone (CRH) and inflammatory cytokines were determined by ELISA.In addition,the research also focused on the changes of CRF-led skin HPA axis systems.CRH/proopiomelanocortin(POMC) axis elements,corresponding functional receptors and crucial genes of glucocorticosteroidogenesis in the skin were measured by quantitative RT-PCR and Western blot.Results: We found that AD mice had a marked reduction in hypothalamic-pituitary-adrenal (HPA) tone by reduced serum CORT and ACTH,and these situations have significantly improved after BSYQ treatment.The AD mice also showed a disturbed peripheral HPA expression.Compared with normal mice,AD lesion skin were detected significantly reduced CRH mRNA and protein expression,on the contrary,POMC and cytochrome P450 side-chain cleavage enzyme (CYP11A1)were significantly increased,meanwhile,Nr3cl(GR),CRH-R2 and 11-hydroxylase type 1 (CYP 11B 1) were significantly down-regulated in the AD mice locally.Conclusions: Our results collectively suggest that AD mice displayed differential central and peripheral HPA axis tone and BSYQ significantly relieved clinical syndrome of AD mice and presented perfect anti-allergy and anti-inflammatory effects by normalizing central and peripheral HPA axis expression.
