Vitamin A supplementation alleviate extrahepatic cholestasis liver injury via Nrf2/NQO1 pathway
Vitamin A is an essential fat-soluble vitamin with antioxidant properties.Evidence has shown that vitamin A could ameliorate cholestatic liver injury, but the related mechanisms remain to be elucidated.Keapl-Nrf2-ARE signaling pathway plays an important role in the cellular defense against oxidative stress and NQO1 is a highly-inducible antioxidant enzyme that is regulated by the Keapl/Nrf2/ARE pathway.In the present study, the nuclear factor-erythroid 2-related factor 2 (Nrf2) and NADPH-quinone oxidoreductase 1 (Nqol)expression in bile duct ligated rats were investigated by immunohisto-chemistry and western blot analysis.The results revealed that Nrf2 and NQO1 expression were upregulated in liver tissue,oxidative stress and liver injury were improved apparently after vitamin A administration.Therefore,we concluded that the mechanism of protect effects of vitamin A in obstructive jaundice rats may be related to promote the activation of Nrf2 and NQO1.
