Microscopic Structural Comparison of Four Different Species in Aconitum L.Based on Serial Sections
Authentication is the first priority in quality evaluation of Chinese herbal medicines (CHMs), especially for those highly toxic medicines.The most commonly used authentication methods include morphological identification and microscopic identification.Unfortunately, these two methods can not effectively evaluate some herbs with complex interior structures, such as Caowu with circular conical shape and successive interior structure.Therefore, which part should be selected plays an essential role in accurate microscopic identification.In this study, we compared the interior structure of roots and tubers of aconitum using serial sections.This protocol can be applied to a large set of structure for microscopic identification, including cortex, cambium, vessel and pith.Our data showed that for an accurate comparison and identification, the cross section should be selected from the root part with the maximum diameter due to its stable and successive interior structure.
