Preparation,characterization and mechanism study of goblet cell-targetingmulti-force crosslinked nanoparticles and the influence of mucus
Multi-force crosslinked trimethyl chitosan nanoparticles (M NPs) with the combination of multi ion electrostatic attraction, macromolecule entanglement and hydrophobic interaction and their goblet cell-targeting nanoparticles (M-CSK NPs) were prepared and characterized.Compared with single ion crosslinked nanoparticles (S NPs), in combined with the stability and targeting effects, these nanoparticles led to the enhanced insulin uptake and transport across the Caco-2/HT29-MTX co-cultured cells via clathrin, caveloae-mediated endocytosis and macropincytosis.Besides, both multi-force nanoparticles were able to transiently open the epithelial tight junctions among cell monolayers, which involving JNK-dependent pathway.The mucus was found to facilitate the drug transportation of both multi-force NPs.Finally, the in vivo studies in diabetic rats demonstrated that M-CSK NPs produced a stronger and more sustained hypoglycemic effect than those of S-CSK NPs and M NPs.
