S-allylcysteine (SAC),A Garlic Derivative, Suppresses Proliferation and Induces Apoptosis of Ovarian Cancer Cells

  ”Aim” Garlic-derived compounds have been widely used for their medicinal purposes to overcome the poor prognosis of ovarian cancer.In this study,we investigated the antiproliferative and apoptosis-inducing effects of S-allylcysteine (SAC), a water-soluble garlic derivative, on human ovarian cancer cell line A2780.”Methods” Colony forming assay,Hoechst 33258 stain,Annexin V/PI staining, transwell invasion,wound healing and western blot assays were performed to analyze the SAC treated A2780 cells.”Results” We found that the proliferation of A2780 cells was inhibited and the apoptosis of A2780 was induced by SAC in a time-dependent manner.In addition,SAC remarkably reduced the migration of A2780 cells.Meanwhile, SAC potently downregulated the protein expression of pro-caspase 3,Parp-1 Bcl-2 and upregulated the expression of active-caspase 3 and Bax.Furthermore,SAC remarkably decreased the protein expression of Wnt5a,pAKT,c-Jun,which were key proteins in proliferation and metastasis.”Conclusion” SAC,as a promising naturally-derived drug candidate, suppresses the proliferation and induces apoptosis of ovarian cancer A2780 cells.
