Molecular Characterisation and Fibrolytic Activities of Neocallimastix and Orpinomyces Fungi Obtained From the Rumen Contents of Wild Yaks (Bos Grunniens)
Fourteen fungal isolates were obtained with a Hungate”s roll-tube technique from rumen contents of wild yaks (Bos grunniens) and they were identified into fungal groups of the monocentric genus Neocallimastix and the polycentric genus Orpinomyces based on their morphological characteristics and internal transcribed spacer region 1 sequences.Acetyl esterase (AE, 168.15 vs.140.49 mU), ferulic acid esterase (FAE, 4.94 vs.3.44 mU) and xylanase (658.36 vs.620.28 mU) activities were higher in the Neocallimastix group than in the Orpinomyces group.Enzyme activity of FAE was highly correlated with activities of AE (r=0.79, P<0.0001) and xylanase (r=0.65, P=0.0008) while the lowest coefficient occurred between AE and xylanase (r=0.52, P=0.0051).In brief, anaerobic fungi exhibited higher fibrolytic activities in the genus Neocallimastix than those activities in the genus Orpinomyces in the rumen contents of wild yaks, suggesting these fungi might play an important role in fibre degradation in the rurnen.
