Interferon-α suppresses invasion and enhances cisplatin-mediated apoptosis and autophagy in human osteosarcoma cell

  Interferon-(IFN-α) is generated in response to viral infections and used clinically in the therapy of a variety of viral infections and cancers.We investigated that IFN-α could inhibit the invasive ability of osteosarcoma cells by Matrigel invasion assay.Then, we treated osteosarcoma cells with cisplatin and/or IFN-α.apoptosis and autophagy were assessed by MTT assay, Hoechest33258 staining ,flow cytometry assay, Acridine orange staining, GFP-LC3 dotted assay and Transmission electron microscopy.Further analysis showed that the effective of cisplatin could be enhanced by combining it with the cytokine IFN-α.These results indicated that the combination therapy of chemotherapeutics and IFN-α is a new way for osteosarcoma which is needed further proved by the experiments in vivo.
