Optimization of Microwave-Assisted Saponification (MAS) for Simultaneous Determination of 1-Octacosanol and 1-Triacontanol in Beeswax by Fast Gas Chromatography

  Determination of contents of 1-octacosanol and 1-triacontanol in natural wax of plant and animal origin are essentially important for the study of their beneficial physiological activities and health benefits.In this paper, a robust, fast and effective analytical method for simultaneous determination of 1-octacosanol and 1-triacontanol in beeswax was established and validated based on fast gas chromatography combined with flame ionization detector (FID).A sample pretreatment coupled with microwave-assisted saponification and non-derivatization prior to analysis has been optimized.Compared with conventional GC and traditional saponification, the proposed method was less time-consuming, more effective and more environmentally friendly due to involving an HP-1MS fast capillary column and microwave irradiation and non-derivatization step.The method had a good linearity over the range 1-500 mg/L with good linear regression correlation coefficients above 0.997, and showed an excellent intra-day and inter-day precision (RSD≤3.8%).The recovery was 86.5%-110.2% (RSD≤5.04%).The limit of quantitation (LOQ) (S/N =10) of this method was determined to be 5 mg/L and 8 mg/L for 1-octacosanol and 1-triacontanol, respectively.
