Comparison Between RAPD and AFLP Markers in Analyzing the Genetic Diversity of Cultivars and Its Application in Potato Breeding

  The genetic diversity of Kexin series potato cultivars, and the application of RAPD and AFLP markers to the practice of potato breeding were studied using two methods.Seven primer combinations were used in the analysis of AFLP, and 54-90 bands were amplified in each lane, whereas 4-9 bands were amplified by each RAPD primer.In RAPD, the genetic distance of Kexin potato cuitivars was between 0.1707-0.7222 and the average value was 0.3917.In AFLP, the genetic distance was between 0.2091-0.7679 and the average value was 0.4811.The Shannon-weaver index and Simpson index from AFLP were all higher than those from RAPD, indicating that the genetic diversity identified by AFLP was higher than that by RAPD.The cluster analysis based on RAPD and AFLP showed that Kexin series potato cultivars could be divided into three groups.Both markers proved in the molecular level that the genetic basis of the Kexin series potato cuitivars was wider.From the results, it was observed that: RAPD was more suitable to detect the genetic distance and analyze the kinship of potato cultivars because it was a simple, fast, and low-cost technique.However, the AFLP markers were more suitable to reveal the difference of potato cultivars and identify the important breeding resource materials.
