A Novel Animal Model of Impaired Glucose Tolerance Induced by the Interaction of Vitamin E Deficiency and Radiation

  OBJECTIVE To evidence two factors,VE deficiency in diet and exposure to radiation,which are commonly encountered in daily life,possibly induce oxidative stress,and to establish a new animal model of impaired glucose tolerance (IGT).METHODS SD rats were fed high lipid and sucrose diet and treated with individually or in combination of two factors,VE deficiency and radiation.SOD,MDA and AGEs were evaluated,respectively.Blood sugar and serum insulin were detected to calculate insulin sensitivity index.To investigate IGT,intravenous glucose tolerance test was conducted.Finally,the insulin level in perfusate from pancreas was examined.RESULTS Oxidative damages were observed in rats of the radiation and VE deficiency treated groups.In the end,IGT was observed only in the group with both factors.Accidently,an interactive effect between two factors was found.Insulin secretion level response to glucose characterized by a rapid but reduced first phase and an obviously defective second phase upon pancreas perfusion under VE deficiency and radiation.CONCLUSION The interaction of VE deficiency in diet and exposure to radiation could break the equilibrium of oxidation and antioxidation,and resulted in IGT.More importantly,a new IGT model was successfully established.
