Dissection of Mechanism of a Chinese Medicinal Formula,Danhong Injection Therapy for Myocardial Ischemia/Reperfusion Injury in Vivo and in Vitro
OBJECTIVE To investigate the primary active ingredients of Danhong Injection (DHI) and their systemic and interrelated mechanism in an established myocardial ischemia/reperfusion (MI/R) model.METHODS The fingerprint was established to analyze the active ingredients.The IL-1,IL-10,TNF-α,MDA and SOD levels in the MI/R rat myocardium and primary cultured cardiomyocytes was detected by ELISA.Flow cytometric analysis and TUNEL assay was used to detected apoptosis rate.Western blot was used to test the expression of key molecular on the signal pathway.RESULTS Hydroxysafflor yellow A (A),salvianolic acid B (B),and danshensu (C) were the main active constituents in DHI.Compound A exerted a more prominent anti-inflammatory effect than B and C,by decreasing NF-κB levels; compound B had more powerful anti-oxidative capacity than A and C,by increasing Nrf2 expression; and compound C had stronger anti-apoptotic ability than A and B,by lowering caspase-3 activity.These compounds and DHI increased phosphorylation of Akt and ERK1/2 in cardiomyocytes.CONCLUSION The main active ingredients in DHI were hydroxysafflor yellow A,salvianolic acid B,and danshensu,which exert anti-inflammatory,anti-oxidative,and anti-apoptotic ability against MI/R induced injury,respectively.
