Comparative evaluation two transdermal preparations on permeation and toxicity:in vivo and in vitro
The aim of this work is to comparative evaluation of two transdermal preparations on permeation and toxicity of microemulsion and gel containing Flurbiprofen: in vivo and in vitro.Skin permeability using a modified Franz diffusion cells in vitro.The flux from ME and gel were 40 and 30 μg·cm-2·h-1 (p< 0.05),respectively.In vivo retention study,ME showed higher permeation amount than gel at the later stage.It reached the peak concentration at 270 ug/ml,then decreased until 40ug/ml.The skin treated with the ME was free of gross damage.Gel shows higher damage on skin than ME and normal saline.In DSC test,the lipid transition at 50 °C disappeared from gel thermograms,while in ME thermogram,the peak at 50 °C did not disappear completely but showed smaller peak area than the control.Both ME and gel of the lipid transition peak of 130 °C deceased to 120°C with reducing of endothermic peak area to some extent,especially for gel.Compared with the two formulations,the present study illustrates ME shows high transdermal permeation and low toxicity.These results suggest that ME is suitable for topical application.
