The anti-hyperuricemia and xanthine oxidase inhibition effects of chrysoeriol, a component from Apium graveolens L.seeds
In this study,twenty-one compounds were isolated from the ethanol extract of Apium graveolens L.,which showed an anti-gout activity in rat model.To screening a potential agent for anti-hyperuricemia and xanthine oxidase (XOD) inhibitor from the bioactive extract,the generation of uric acid method and the NBT reduction method were used in vitro.The results demonstrated that chrysoeriol was a strong XOD inhibitor (IC50=5.6 μM) with a competetive inhibition effect.To further evaluate the anti-hyperuricemia effect of chrysoeriol,the reducing serum uric acid levels in oteracil potassium induced hyperuricemia mice by chrysoeriol was further investigated.The hyperuricemic mice were oral administrated chrysoeriol at a dosage of 10mg/kg,20mg/kg and 30mg/kg,the uric acid values in serum were all reduced significantly.Based on the results obtained in this study,chrysoeriol may be a potential lead compound for developing the pharmaceutic for anti-hyperuricemia agent.
