Pharmacognostic investigation of Ardisia primulaefolia and its effect on HeLa cancer cells
Aim of the study: In the context of the present study,the chemical composition and relative anticancer potential of A.primulaefolia was investigated,and the detailed pharmacognostical evaluation of A.primulaefolia has been carried out with the aim to establish the identification markers of this important antitumor drug used commonly in TCM and provided referential information for research of this genus.Materials and methods: The A.primulaefolia was collected in November 2010 from Guangxi province of China and the plant was authenticated.Crude extract of A.primulaefolia was prepared in 85% EtOH under reflux and was fractionated into two organic and one aqueous fraction on polarity basis.Cytotoxicity on cervix adenocarcinoma cell line (HeLa) was carried out in vitro in two organic and one aqueous fraction of A.primulaefolia.The pharmacognostic profile,physicochemical parameters of the Ardisia primulaefolia were carried out.Macroscopic and microscopic characteristics as well as transverse section of the A.primulaefolia were studied.Results: Among the three extracts tested,HeLa cell line using MTT assay showed a percentage of cellviability of 85.6% at 50μg/ml which decreased with increase in concentration of n-butanol fraction,followed by aqueous fraction.Alcohol extraction had effective antitumor potential.Cytotoxicity (IC50 = 32.9μg/ml) against HeLa cancer cells A.primulaefolia contains various compounds such as saponins,quinones,tannins,flavonoids etc.It is particularly rich in saponins.The crude ethanol extract and its subsequent solvent fractions were tested for their physicochemical parameters such as ash values,moisture contents and extractive values were carried out.Conclusions: Although the mechanism is not entirely clear,are the three fraction of A.primulaefolia with the greatest anticancer potential against HeLa cells in vitro and deserve further investigation.The results obtained provide the basis for isolation and identification of biologically active substances from the three fractions.The results of this research establish parameters for the standardization which will be helpful for A.primulaefolia used commonly in TCM.This study also could give prevention from adulteration for the plant identification in TM.
