Effect of piperine on the pharmacokinetics of curcumin
Purpose: The effect of single and multiple doses of piperine on the bioavailability and pharmacokinetics of curcumin was studied in healthy volunteers.Methods: Nine healthy volunteers aged 20–25 years were randomized to receive placebo or 20 mg of piperine daily.After seven days,the single-dose pharmacokinetic profile of an 600-mg dose of curcumin was evaluated.Study participants received each of three study regimens in one of three randomly assigned orders.Blood samples were collected,and plasma curcumin concentrations were analyzed using liquid chromatography (LC)/tandem mass spectrometry (MS/MS) method.Results: The coadministration of single-dose of piperine with curcumin increased the mean curcumin area under the concentration-versus-time curve (AUC0–60h) by 17% compared with placebo.The tmax and t1/2 increased by 44 and 47%,respectively whereas CL/F decreased by 33%.Prior administration of multiple-dose of piperine produced an increase of 65% in AUC0–60h and 42% in Cmax; The tmax and t1/2 prolonged by 80 and 55%,respectively,while CL/F decreased by 51%.However the difference in pharmacokinetic parameters of curcumin among the three phases was not statistically significant(p>0.05).Conclusion: The present study demonstrates that co-administration either of single or multiple doses of piperine failed to enhance the curcumin bioavailablity according to the ANOVA.This results are unlikely to be clinically relevant.Further in-depth studies in a large number of participants are required to illuminate precisely its interaction with curcumin.
