Preventative effects of Ginkgo biloba extract (EGb761) on high glucose-cultured opacity of rat lens
Aim: To investigate the protective effects of EGb761 on high glucose-induced opacity of rat lens.Methods: After rat lenses were excised from the eyes, lenses were divided into 6 groups: normal group, high glucose group, low-, moderate-, and high-dose of EGb761 groups, and bendazac lysine group.The activities of antioxidases, aldose reductase (AR), advanced glycosylation end products (AGEs), transforming growth factor-β2 (TGF-β2), Smad2/3, E-cadherin, and α-SMA were assessed.Results: Compared with in high glucose group, EGb761 relieved oxidative stress, decreased AR activation and AGEs level, and suppressed the TGF-β2/Smad pathway activation, further increase the expression of E-cadherin and decrease α-SMA, and therefore prevents the pathological changes of high glucose-induced lens epithelial cells and ameliorated lens opacity.Conclusion: EGb761 has protective effects on several pharmacological targets in the progress of diabetic cataract and is a potential drug for the prevention of diabetic cataract.
