Depletion of Interleukin-8 expression through RNAi induces reversal of BCNU resistance in glioma
Our previous study had cloned two glioma cell lines SWOZ1 and SWOZ2 isolated from parental glioma cell line SWO38.The 1,3-bis(2-chloroethy1)-1-nitrosourea (BCNU) resistance of SWOZ1 were higher than that of SWOZ2.In our further research,the glioma cell line SWOZ2-BCNU,which is resistant to BCNU compared to parental cell line SWOZ2,was established.Interestingly,interleukin-8 (IL-8) was found to be both upregulated in BCNU-resistant cell lines including SWOZI and SWOZ2-BCNU.To further investigate the function of IL-8 in glioma resistant to BCNU,siRNAs targeting IL-8 were synthesized and transfected into the cell lines SWOZ1 and SWOZ2-BCNU.After treated by siRNA,s targeting IL-8,a depletion level of IL-8 mRNA expression in cell lines SWOZ1 and SWOZ2-BCNU for more than 90% compared to negative control was found and confirmed by western blot(P<0.05),with a reversal BCNU-resistance for about 50% both in these two BCNU resistant cell lines (P<0.05).Our data demonstrated that silencing IL-8 with specific siRNAs can reverse the BCNU resistant phenotype in these glioma cell lines,which indicate that IL-8 may play.an important role in glioma resistant to BCNU.
