IDH mutation Improves the Prognosis in Chinese Glioma Patients with TP53 Over-expression

  Objective:To investigate the mutational status of isocitrate dehydrogenase 1 (IDH1) and IDH2 of 109 Chinese glioma patients.Methods:Total of 109 Chinese glioma patients was involved in this study.The mutational status of IDH1 and IDH2 of 109 Chinese glioma patients was investigated using immunohistochemical analysis and DNA sequencing.The status of TP53 expression was also investigated using immunohistochemical analysis.Formalin-fixed,paraffin-embedded 5-μm thick tumor tissue sections were used for both DNA isolation and immunohistochemical staining.Anti-human IDH1 R132H and anti-TP53 primary antibodies were used to detect the IDH1 codon 132 mutation (R132H) and TP53 expression,respectively.IDH1 and IDH2 PCR products were sequenced to confirm the presence of IDH1 R132 or IDH2 R172 mutations.
