Influence of High Concentration Tetrahydrofuran on MetabolicProfiling of Rhodococcus sp.YYL Using H-NMR Spectroscopy andMultivariate Data Analysis
Although tetrahydrofuran (THF) is a toxic and carcinogenic compound and has toxicity effects to organisms metabolism,Rhodococcus sp.YYL is known asutilizing THF as the sole carbon source and energy for growth.The genetic mechanism of high concentration THF stress survival for Rhodococcus sp.YYL containing tetrahydrofuran monooxygenase gene has been elucidated; however,the downstream metabolic effects have not yet been evaluated.In this study,we determined the metabolic profiling of Rhodococcus sp.YYL being cultured with onlylg/L yeast extract (CY),with 20 mM THF and lg/L yeast extract (TY),and with only 20mM THF (CT) throughout a time course (48h and 84h) using 1H-NMR spectroscopy and multivariate data analysis.We found that the dominant metabolites in Rhodococcus sp.YYL were two sugars,seven amino acids,five aliphatic organic acids,six nucleotides and three amines.
