Influence of cell disruption and elution on cellulase release of Clostridium straminisolvens CSK-1
CSK-1 is the only cellulose-degrading bacterium that has been isolated from bacterial community MC1.Release of the cellulase from CSK-1 cells is a challenging step for its research of cellulase characteristic and further function mechanism.In this study,the influence of cell disruption and elution on cellulase release of CSK-1 was investigated by determining five kinds of cellulase activity.Six different methods were used for raw sample treatment:freezing-thawing,ultrasonication,elution,freezing-thawing after elution,ultrasonication after elution and homogenization high pressure after elution.The results showed that high pressure homogenization after elution was the optimal method for endoglucanase,exoglucannase and hemicellulase release,the corresponding enzyme activity increased by 81.5%,87.7% and 51.7%;Ultrasonication before elution was the optimal method for glucosidase release and the enzyme activity increased by 23.8%; Freezing-thawing before elution was the optimal method for filter paper enzyme release and the enzyme activity increased by 46.3%.
