Haematococcus pluvialis-Based High-throughput Screening for Enhanced Activity of Atrazine Chlorohydrolase by Directed Evolution
The atrazine chlorohydrolase (AtzA) attracts widespread interests as it catalyzes the toxic atrazine to non-toxic hydroxyatrazine and can be used in the biodegradation of atrazine.To facilitate this application,we developed a Haematococcus pluvialis-based high-throughput method to screen AtzA mutants from random mutagenesis libraries.Eight variants with enhanced enzyme activity were successfully obtained.All variants showed significant increase in their specific activity,ranging from 2.2-to 5.1-fold compared to the wild-type.
