CTGF receptor avβ3 integrins play an important role in myofibroblastic differentiation of hypertrophic scar fibroblasts

  Connective Tissue Growth Factor (CTGF) is a potent mediator of the differentiation of hypertrophic scar fibroblasts into myofibroblasts,which is characterized by the appearance of the cytoskeletal protein a-smooth muscle actin.The aim of this study was to investigate the role of avβ3 integrins in CTGF-induced myofibroblast differentiation.We show that CTGF induces hypertrophic scar fibroblasts differentiation and collagen synthesis in mRNA and protein level.In this study,we compared the levels of av、β3 integrins between atrophic scar and hypertrophic scar and investigated the association with hypertrophic scar.We demonstrated that av integrins was up-regulated in hypertrophic scar.The expression of av integrins mRNA and protein was up-regulated by TGF-β1 stimulation during the myofibroblasts transition.Then,blockade of the avβ3 integrins prevents the CTGF-induced myofibroblast differentiation in hypertrophic scar fibroblasts.These data identify a novel mechanism for the establishment of autocrine CTGF signaling in hypertrophic scar fibroblasts by the up-regulation of CTGF receptor avβ3 integrins and suggest the possibility of regulating fibrotic disorders,especially hypertrophic scar,by targeting this integrin.
