Short sequence and secondary structure of ITS2, compared to the full length ITS, provide efficient taxonomic sequence tags
An ideal DNA barcoding region should be short enough to amplify from degraded DNA.In this article,we discuss the possibility of mini nuclear DNA sequence in identifying plant species.Firstly,the PCR and sequencing recovery of ITS and ITS2 were evaluated based entirely on materials from dry medicinal product and herbarium voucher specimens with some samples collected 90 years ago.The results showed that ITS2 could recovered 91% while ITS only 23% efficiency of PCR and sequencing by using one pair of primer.Secondly,the ITS and ITS2 sequences of 12862 plant samples including 8303 species,were downloaded from GenBank used for comparing the identification efficiency.
