Rapid determination 24 kinds of amino acids in the Pollen of Ginkgo biloba by UPLC-TQ-MS
Objective:To establish a method for rapid determination 24 kinds of amino acids in the pollen of Ginkgo biloba without derivatization,and apply it in the determination of pollen,anther,stamens and flower spike of Ginkgo biloba.Method:Determin it by UPLC-TQ-MS,The HILICcolumn utilized was an Acquity UPLC BEH Amide (2.1 mm × 100 mm,1.7 μm).The mobile phase consisted of A (water,5 mmol/l ammonium format,5 mmol/L ammonium acetate and 0.15% formic acid) and B (acetonitrile,1 mmol/L ammonium format,1 mmol/L ammonium acetate and 0.15% formic acid),and was used in gradient elution.UPLC linear gradient conditions were:0-6 min,15-20% A; 6-10 min,20-30% A; 10-11 min,30-40% A.The flow rate of the mobile phase was 0.4 ml/min.Result:The 24 kinds of amino acid had the respective concentration linear range (r2 > 0.9899);recoveries were 93.1%-103.6%,RSD 1.04~8.95%.The total amino acids of pollen> anther>stamens>flower spikes of Ginkgo biloba,Essential amino acid content of pollen =anthers> stamens> flower spikes.Results:.This method has a good accuracy and precision.The amino acids of pollen are more than 6.43%,the amino acids in anther,stamens and flower spikes are also high.The analysis of results provide basis for the development of health products of pollon of Ginkgo biloba.It can also provide reference for the comprehensive development of anther,stamens and flower spike of Ginkgo biloba and save resources.
