Protein Microarray Analysis in a Mouse Cecal Ligation and Puncture Model for Early Prediction of Sepsis-Induced Disseminated Intravascular Coagulation
Objective To identify valuable biomarkers of sepsis-induced disseminated intravascular coagulation (DIC) from platelets-derived factors by using a biotin label-based custom protein microarray technology in a mouse cecal ligation and puncture (CLP) model.Methods KMmice were randomized into sham-operated group and CLP group.Blood were detected immediately,at 1hr,2 hrs,6 hrs,12 hrs,24 hrs,48 hrs and 72 hrs after CLP for platelet count,coagulation assay and blood chemistries.Histological assessment of the lung and mesenterium tissue was executed at all corresponding time points for microthrombus formation.Serial detection by protein microarray analysis was performed for platelets-derived factors.Results The survivor probability at the time point of 72hrs post-CLP was 15% and there was no mortality among the sham mice.
