Simultaneous Determination of Naphthoquine phosphate, and Azithromycin in Human Plasma by HPLC-MS-MS
A rapid,sensitive and specific assay method has been developed to simultaneously determine human plasma concentrations of naphthoquine and azithromycin,using high-performance liquid chromatography with an electrospray ionization(ESI) tandem mass spectrometry (HPLC-MS-MS).Chromatographic separation was performed on an Agilent-Zorbax-Eclipse-XDB-C18 column(75mm×2.lmm,3.51μm,Agilent,USA) using a gradient mobile phase with ultrapure water containing 0.1% formic acid and methanol by gradient elution.Detection and quantitation were performed by MS/MS using electrospray ionization and multiple reactions monitoring in the positive ion mode.The calibration curves were linear over the concentration ranges 0.25-100 ng/mL for naphthoquine (r=0.9990) and 1-1000 ng/mL for azithromycin (r =0.9974).The limit of quantification was 0.25ng/mL for naphthoquine and 1 ng/mL for azithromycin.The relative recovery was 99.97±4.89,96.97±3.52 and 100.02±2.27% for naphthoquine,110.13±7.83,88.42±1.67 and 102.13±2.22% for azithromycin.The method was accurate,sensitive and simple and was successfully applied to a pharmacokinetic study after a single oral administration of the combination at a dose of 400 mg in 12 healthy Chinese volunteers.
