Quantification of exenatide in human plasma using high-resolution/accurate mass LC-MS/MS
Objective:Exenatide is a polypeptide containing 39 amino acids.It”s a synthetic glucagon-like peptide (GLP-1)analog,and has a high affinity to GLP-1 receptor,clinically used for the treatment of type 2 diabetes.Conventional analysis method for quantification of exenatide is immunoassay technique,but their specificity,stability and reproducibility is poor,and the method is expensive.LC-MS is a rapidly development method for drug analysis in biological matrix in recent years.However,the low plasma concentration of exenatide and interference from endogenous substances bring a huge challenge for the evaluation of its pharmacokinetic characteristics.Triple TOF is a new mass spectrometer with high-sensitivity,high-resolution and high-quality accuracy under maximum scanning speed.It”s able to provide similar MRM quantitative ability of high-resolution mass spectrometry,the lowest limit of detection and dynamic range can be quite high-end triple-quadrupole mass spectrometry system.
