Expression of Synaptophysin and Its mRNA in Bovine Corpus Lutea During Different Stages of Pregnancy
In order to investigate the expression of mRNA and protein for synaptophysin in bovine corpus lutea during different stages of pregnancy,we chose Holstein cows during various pregnancy stages,and the corpus luteum was divided into two parts,then immunohistochemical streptavidin-perosidase and RT-PCR were used to determine the levels of protein and mRNA for synaptophysin.Synaptophysin immunoreactive products mainly located in large luteal cells,much less or no immunoreactivity was found in small luteal cells and other parts of the ovary.The expression levels of synaptophysin were different in various stages of pregnancy.In mid-pregnancy,the number of positive cells was high and stained stronger than that in early pregnancy (P<0.05).The number decreased and the expression was weak in late pregnancy (P<0.05); and the number of positive cells reduced quickly after delivery (P<0.05).The synaptophysin mRNA expression in mid-pregnancy was significantly higher than that in early and late pregnancy (P<0.05),and the results were consistent with that of immunoreactivity.We concluded that there was a difference in the expression of synaptophysin at various stages of pregnancy,indicating that synaptophysin was closely related to the luteal function and reproductive regulation.
