Tongxinluo reverses metabolic abnormalities,and the structure and function of the arteries of hypertensive rats with metabolic disturbances(Abstract)
Hypertension,dyslipidemia,and hyperglycemia have key roles in most life-threatening diseases(e.g.,coronary artery disease,diabetes mellitus,cerebral apoplexy).Effective therapies for these metabolic disturbances are lacking.Herbal medicines may be suitable for management of poly-metabolic disturbances due to their multiple target effects.This study aimed to evaluate the effect of a compound herbal medicine,Tongxinluo(TXL),in a rat model of metabolic disturbances(MD) from spontaneously hypertensive rats(SHR) fed a high-cholesterol diet.TXL at low(0.2 g/kg) or high dose(0.8 g/kg) and simvastatin(1.2 mg/kg) was administered for4 weeks starting from week 5 after the start of a high-cholesterol diet.Rats were monitored with regard to cardiovascular and biochemical parameters at week 4 and week 8.Feeding of a high-cholesterol diet resulted in higher fasting blood glucose and c-reactive protein(CRP);impaired glucose tolerance;and elevated total cholesterol,LDL and HDL.Treatment of MD rats with TXL for 4 weeks resulted in reduced thickness of the right common carotid artery,reduced CRP,improved glucose tolerance and heart function.Ex-vivo studies in isolated artery rings demonstrated that TXL pretreatment resulted in significantly improved function of endothelial cells and smooth muscle cells of the aorta.Treatment of TXL resulted in a significanfly increased aortic peroxisome proliferator-activated receptor-mRNA expression compared with control SHRs.TXL is a promising herbal preparation with multiple targets in management of the metabolic syndrome.
