Selection of Equivalent Waveband and Discrete Wavelength Combination for Hemoglobin Measurement with VIS-NIR Spectroscopy
Visible and near infrared(VIS-NIR)spectroscopy combining with the improved moving window partial least-squares (MWPLS)method was applied to the wavelength selection for reagent-less determination of Hemoglobin(Hb)in human whole blood.A new modeling approach was performed based on randomness,similarity,and stability.Experimental results indicate that the VIS-Short NIR region 400-1100 nm can be used as the information waveband of Hb in human whole blood.Based on the MWPLS method in 400-1100 nm,the optimal waveband was 492-890 nm,and a model space including 77 wavebands with equivalence for the optimal waveband was proposed;the span of 77 equivalent wavebands was 436-898 nm,it contains sufficient information of Hb.In the waveband of 436-898 nm,discrete cornbination multiple linear regression(DCMLR)models were further selected;the optimal model was the MLR model of three wavelengths(544,740,and 754 nm).Validation effects V-SEP,V-Rp,and V-RSEP for the PLS model in 492-890 um were 2.58 g L-1,0.988,and 1.97%,respectively;validation effects V-SEP,V-Rp,and V-RSEP for the MLR model of the three wavelengths were 2.82 g L-1,0.979,and 2.15%,respectively;two models both achieve high prediction accuracy,they may be used in clinic.Various equivalent wavebands and discrete wavelength combinations resolve restriction of position and number of wavelengths in instrument design of discrete and continuous type.
