Heterotopic osteogenesis of spherule accumulating scaffolds with different proportions of hydroxyapatite/alpha calcium sulfate hemihydrate in vivo
Fabrication three different mass ratio of hydroxyapatite/alpha calcium sulfate hemihydrate (HA/α-CSH) compositespherulites by the method of sol-gel (water/oil emulsification) technique. The spherulites were filled into the porous HA tubecoated with polylactic acid and sealed with HA discs to form the porous scaffold. They were implanted in three non-osseous sitesin dogs: the abdominal muscle(AM), peritoneum pocket (PP), and dorsal muscle (DM). Six months after implantation, scaffoldsimplanted in AM and DM degraded with small fractions remaining observable. In comparison, those implanted in PP maintainedthe generally complete shapes. The results of histomorphometric analysis indicated osteogenesis in all scaffolds samples in PPgroups. In addition, the sample composed 75%HA-25%α-CSH induced significantly more bone than the other implants. Theresults demonstrated that in the absence of added growth factor and drug, reasonable porosity and degradation rate wereadvantageous for ectopic osteogenesis and the peritoneum can be used as the better target site for bone tissue engineering in vivo.
