Disulfide Cross-linked Low Generation Dendrimers with High Gene Delivery Efficiency, Low Cytotoxicity, and Low Cost

Cationic poly(amidoamine) (PAMAM) dendrimers were widely used asnon-viral gene carriers. PAMAM dendrimer-based products such as Superfect?And Priofect? Were already commercially available gene transfection reagents.However, these products are based on high generation dendrimers with high costand serious cytotoxicity. In this study, we prepared high efficient gene carriersusing disulfide cross-linked generation 2 (G2) PAMAM dendrimers. Thesedisulfide cross-linked G2 dendrimers degrade into G2 dendrimer after cellularuptake. They can effectively condense DNA into ~200 nm polyplexes. Comparedto G2 and G5 PAMAM dendrimers, disulfide cross-linked G2 PAMAM dendrimershowed much improved gene transfection efficiency (both EGFP gene andluciferase) and reduced cytotoxicity in both 293T and HeLa cell lines. Thedisulfide cross-linked G2 dendrimer prepared at a linker/dendrimer molar ratio of1:1 showed the highest gene delivery efficiency and exhibited comparabletransfection efficiency to branched PEI 25 kD (bPEI 25 kD), a commerciallyavailable non-viral gene vector. Confocal laser scanning microscope (CLSM)studies further revealed the high cellular uptake behavior of disulfide cross-linkedG2 PAMAM dendrimers. Our study have demonstrated that disulfide cross-linkedlow generation dendrimers with low cost, cytotoxicity, and high transfectionefficiency are efficient alternatives to high generation dendrimers in gene delivery.
