Hydrogen sulfide and cystathionine-γ-lyase promote acute lung injury induced by acute pancreatitis in rats

Acute pancreatitis(AP)is well-known to induce the injuries to extrapancreafic organs.Because respiratorydysfunction is the main cause of death in patients with severe AP,acute pancreatitis-associated lung injury(A-PALI)is a great challenge for the clinicians.This study aimed to investigate the potential role of H2S in thepathogenesis of APALI.54 SD rats were radomly divided into three groups:AP group made by injection of sodiumdeoxycholate into the common bile duct,control group made by sham operation’and treatment group made by in—trapefitoneal injection of propargylglycine(PAG.CSE inhibitor)into AP rats.Histopahtology of the lung Was ex—amined and the exDrcssion of CSE and TNF—α mRNA in lung tissue Was detected by RT-PCR.H2S level in thescmm WLq detected spectrophotometrically.The results showed that the serum concentration of H2S and CSE andTNF-α expression in the lung were increased in AP rats.Intraperitoneal injection of PAG could reduce theserum concentration of H2S,reduce CSE and TNF-α expression,and elleviate the lung pathology.Taken togeth-er.our findings suggest that H2S/CSE system is crucially involved in the pathological process of APALI and rep-resents a novel target for the therapy of APALI.Acute pancreatitis associated lung injury;H2S;cystathionine-γ-lyase;propargylglycine;TNF-α
