MALDI—TOF MS—based identification of Salmonel la at species and serotype levels

Salmonella can cause illnesses in humans and many animals.Thecurrent diagnostic gold standard used for detecting Salmonella infection ismicrobi010gical culture followed by serological confirmation tests.However,these methods are compl icated and time—consuming.Matrix—assisted laserdesorption/ionization time-of—flight mass spectrometry(MALDI—TOF MS)analysisoffers some advantages in rapid identification,for example,simple and fastsample preparation,fast and automated measurement,and robust and rel iableidentification up to genus and species levels,possibly even to the strain level.In this study,we established a reference database for species identificationusing whole—cel l MALDI—TOF MS:the database consi sted of 12 obtained main spectrafor the Salmonella culture collection strains purchased from the NationaI Centerfor Medical Culture Collection(CMCC)and American Type Culture Collection(ATCC).These 12 Salmonella culture collection strains belong to 7 serotypes.The database was evaluated using 82 clinical isolates of Salmonella,and partial16S rRNA gene sequencing was used as comparision.With regard to theidentification of Salmonella Typhimurium,the accuracy of identification at theserotype level by using MALDI—TOF MS was significantly higher(67.86%)than theaccuracy of identification by using partial 16S rRNA gene sequencing(28.57%)(X2=8.654,P<0.05):nevertheless,there was no significant differencebetween the accuracy of species—level identification by the 2 methods(96.34%and 100%,respectively)(x2=1.358:P>0.05).This study confirmed the highdiscriminatory ability of MALDI—TOF MS for serotype—level identification ofSalmonella.
