Effect of the intranasal immunized with inactived avian H5N2 influenza virus antigen on the local respiratory tract and immune levels of system in chicken
Chicken were immunized via intranasal with inactivated avian H5N2 influenza virus and adjuvant CpG(CpG adjuvant group) or rIL-2(IL adjuvant group). The results showed that the number of IELs in the trachea and the tracheal fork increased significantly (P < 0.05 or P < 0.01) in CpG adjuvant group and IL adjuvant group compared to control group after intranasal immunization;the number of CD3+ cells in the tracheal, tracheal fork and lung increased significantly (P < 0.05 or P < 0.01) in CpG adjuvant group and IL adjuvant group compared to control group after intranasal immunization;the number of mast cells in the laryngeal, tracheal, tracheal fork and lung increased significantly (P < 0.05 or P < 0.01) in CpG and IL adjuvant group compared to control group after intranasal immunization;the number of IgA and IgG secreting ceils increased significantly (P < 0.05 or P < 0.01) in CpG and IL adjuvant group compared to control group after intranasal immunization;but the changes of IgA and IgG secreting cells in the tracheal, tracheal fork and lung were different from each other, the number of IgA and IgG secreting cells in tracheal fork was most, the level of the SIgA in duodenum content increased significantly (P < 0.05 or P < 0.01) in CpG adjuvant group and IL adjuvant group compared to control group after intranasal immunization;the HI degree increased significantly in CpG and IL immune group compared to control group after intranasal immunization, and that the level of specific antibody IgA increased in CpG and IL immune group compared to control group after intranasal immunization, but the changes was not significant. Whereas the effect on the number of IELs and CD3+T cells and mast cells and IgA and IgG secreting cells and the level of the SIgA in the matter of duodenum and specific antibody IgA in serum was no significant changes, HI degree approach 0, there was no specific AIV antibody in the serum after only intranasal immunization with inactivated avian H5N2 influenza virus antigen. The result prompted that intranasal immunization with inactivated avian H5N2 influenza virus antigen and adjuvant could improve the local respiratory tract and immune levels of system in chicken.
