Determination of lincomycin and spectinomycin residues in meat by hydrophilic interaction chromatography-mass spectrometry
A new analytical method, using hydrophilic interaction chromatography-mass spectrometry techniques, was developed for the determination of lincomycin and spectinomycin residues in meat. The analytes were extracted from sample matrix using phosphate buffer with trichloroacetic acid, then the extracts were cleaned up by ion-pair solid phase extraction (SPE), finally the elution was evaporated and brought to volume( 1.0ml) with acetonitrile. Hydrophilic interaction chromatography (HILIC) was used to obtain the necessary separation of the two antibiotics from interfering compounds. Analytes were detected using ellectrospray ionization tandem mass spectrometry. The present method was validated to a low-end concentration (limit of quantitation) of 0.5μ/kg and 10μ/kg for lincomycin and spectinomycin, respectively. This work proposes a newly high selective, accurate and sensitive method to monitor and confirm the residues of lincomycin and spectinomycin in meat.
