Effects of arctiin on streptozotocin-induced diabetic retinopathy in Sprague-Dawley rats

Diabetic retinopathy (DR) is one of the most common and severe complications of diabetes mellitus (DM). In this study, arctiin was extracted from Fructus Arctii and its chemopreventive effect on streptozotocin (STZ) induced DR in male Sprague-Dawley (SD) rats were examined. In the animal experiment, male SD rats were given intraperitoneal injection of 45mg/kg STZ body wt as initiation. Five groups of 20 rats were treated respectively with intragastric doses of 30/90/270 mg/kg/d wt of arctiin, 100 calcium mg/kg/d wt of dobesilate, and 5‰ CMC-Na during the experiment period. Incidence of DR didn”t differ significantly among the STZ-treated groups. At the end of the 16th week, glycosylated hemoglobin (HBA1c) significantly decreased in all arctiin groups as compared with control group after STZ initiation, and concentration of plasma glucose significantly decreased in 270mg/kg/d arctiin group. HE staining showed that arctiin ameliorated edema and detachment of retina. Immunochemistry results confirmed that arctiin ameliorated VEGF expression in retina. In addition, arctiin could increase viability of retinal microvascular endothelia cells (RMEC) evidently in vitro. These findings provided important information of arctiin on ameliorate the severity of diabetic complications, thereby establishing a basis for this herb to inhibit the development of DR and further clinical application.
