Development and evaluation of an HPLC tandem mass spectrometric assay for the simultaneous determination of 25-OH vitamin D3 and D2
25-hydroxyvitamin D(D3/D2)is frequently tested in clinicaflaboratories due to its importance in the management of patients with bone disease and the association between 25-hydroxyvitamin D(25(OH)D)deficiency/insufficiency and certain cancers,autoimmune disorders,infectious diseases,cardiovascular diseases,and many other conditions.Current methodoIoaies for 25(OH)D testing in clinicaI Iaboratories include radioimmunoassay,enzyme-linked immunosorbent assay,high-performance liquid chromatography(HPLC),HPLC coupled mass spectrometry(LC-MS),and chemiluminescent immunoassays.Most Lc-MS methods for 25(OH)D testing are home-brew assays;therefore,carefuI evaluation iS needed prior to clinical application.The objective of this study iS to develop and evaluate an Lc tandem MS(LC-MS/MS)assay for simultaneous quantification of 25(OH)D3/D2.Serum,standards,or controls were mixed with methanol—isoproponoI containing internaI standard(IS,25(OH)D3一d6),and ZnS04 reagent was added followed by vortex mixing. Then methan01 was added and mixed to precipitate proteins.Hexane was added to extract the 25(OH)D3/D2.After centrifugation,the hexane Iayer was dried under nitrogen.
