Molecular engineering of enzymes to serve as ELISA labels for the enhanced quantification sensitivity:a future direction
Enzyme-linked immunosorbent assay(ELISA) iS routinely used for the selective assay of a hapten, all antigen(Ag),or an antibody(Ab)in specimens. The quantification sensitivity of ELISA is by no means satisfactory,however.The principal external factors affecting ELISA sensitivity include the number of an enzyme conjugated to all Ab,the catalytic eHlclency ot tlae conjugate,me reacuon period of the alzyme on its substrate and the sensitivity to quantify enzyme product(s).For practical efficiency,the reaction period is limited.For the reactivity to all analyte of interest,the number of the enzyme conjugated to the Ab is linlited.Therefore.the catalytic efficiency of alzynle·Ab conjugates,together with the sensitivity to quantify enzyme products,should be improved primarily.
