Effect of serm on bovine somatic cell nuclear transfer without micromanipulators
Nuclear transfer (NT) is a complex procedure that requires considerable technical skills. Micromanipulators have been regarded as indispensable for somatic cell nuclear transfer. A significant step forwards has been the development of the so-called handmade cloning (HMC) methods.lt does not require micromanipulators because the manipulations required for both enucleation and nucleus transfer are performed by hand. The purpose of the present work was to optimize certain steps of the micromanipulator-free procedure. Firstly, zona pellucida removal was used by pronase, addition of serum to 15% PBS inhibited the enzyme and decreased the oocytes lysed. Secondly, the survival rate of the bisection medium supplementation in 10% FBS was still significantly higher than those in groups 15% and 20%, individually (P>0..05). Thirdly, the cytoplast-somatic cell attachment is not necessary high concentrations of FBS, 2% FBS is better. The improved handmade somatic cell nuclear transfer method may become a useful technology as a simple, inexpensive, and efficient alternative to traditional somatic cell nuclear transfer.
