Ezpression and Purification of Recombinant Bovine Neuropeptide Y in E.coli
Neuropeptide Y is a 36-amino acid peptide synthesized mainly in the hypothalamus and is one of the most potent orexigenic factors. To explore a approach for high expression of soluble NPY in Escherichia coli cells, we fused the sequence encoding bovine NPY gene to construct an expression vector pET32a(+)-bNPY. The fusion protein hexa-his-trx-bNPY was purified successfully by HisTrap HP affinity column. Immunohistochemical assays demonstrated that NPY was localized in cellular cytoplast and cellular membrane of neurons in cattle hypothalamic arcuate nucleus and the expressed bNPY protein obtained by splicing the gene in vitro can return to the cattle body itself. The procedure described in this study will provide a reliable and simple method for production of NPY to be used in animal production.
