Effect of starfish sterol screened by human arrhythmic related gene on ezperimental atrial fibrillation
To study the effectt of modified starfish steroid C03 screened by human arrhythmic related gene Kv1.5 on experimental atrial fibrillation. METHODS: Kv1.5 cDNA was translated into mRNA and microinjected into Xenopus oocyte. Models of atrial fibrillation was induced by electricity and drug. RESULTS: Pure and single IKur K+ current was expressed on oocytes, without other current pollution. Active material starfish steroid C03 blocking IKur current was screened by patch-clamp . C03 has demonstrated significantly and dose-dependently, to increase AFT induced by electricity in cats, AFT of C03 34.6mg ·kg-1 increased from (4.78 ±0.94)V to (10.87±2.38)V (P<0.01), which is better than Amio; shorten time of improving and recovering induced by Aco, C03 2.7, 8.1 and 24.2mg ·kg-1 shortened the improving time (P<0.01). C03 8.1 and 24.2 mg ·kg-1 shortened the recovering time (P<0.01); shorten time of improving and recovering induced by Ach, C03 8.1 and 24.2 mg ·kg-1 shortened the improving time (P<0.05),C03 2.7, 8.1 and 24.2 mg ·kg-1 shortened the recovering time (P<0.01). CONCLUSION:Starfish sterol C03 produced a oharmacological effects against experimental atrial fibrillation. The model of application of arrhythmic related gene could be used for screening of class III antiarrhythmic drugs from ocean active materials.
