丙型肝炎病毒基因组3”非编码区与细胞蛋白U2B作用的研究

摘要：

To identify the cellular factors interating with HCV 3”NCR RNA，we screened the T7 phage human hepatocyte cDNA libraryand 5 clones were selected including U2B. We used recombinant siRNA vector to silence U2B specifically and then monitored itseffects on HCV replication and translation by quantitive RT PCR and westen blotting respectively. We found that U2B might play anegative role in HCV RNA replication and translation. In addition，we isolated the subGellular fractions of HCV replicon cells and Huh-7cells，and western blot analyse showed that U28 could distribute in the cytoplasm and lipid raft membrane of HCY replicon cells. Ourresults suggest that U28 may be a component of the HCV replication complex.
