Role of FOXO1-Mediated β2-AR/NF-κB Signaling Pathway in Restraint Stress-induced Reproductive Disorder in Pregnant Mouse
The mechanism of mental illness caused by restraint stress(a psychological stress model)during pregnancy is focused by current research.The study mainly aimed to explore pathway by the membrane receptor and intracellular signaling molecules of abnormal hormone under restraint stress in different gestational ages.Restraint stress augmented blood corticosterone(CORT),norepinephrine(NE),blood glucose and decreased estradiol(E2).The increase of NE promoted expression of its receptor β2-AR in uterus and ovary.As well as,the expression levels of FOXO family were increased in uterus and ovary under restraint stress,which elevated ROS levels with decrease of uterine antioxidant capacity(GSH-Px,T-SOD and T-AOC)but increase of MDA.Meanwhile,FOXO can regulate the NF-κB expression when the β2-AR was activated.In conclusion,restraint stress disturbed the stress hormone and E2,triggering the FOXO1-mediated β2-AR/NF-κB signaling pathway to decrease the levels of anti-oxidative.This study laid the foundation for the mechanism study of psychological stress during pregnancy.
